Understanding DMPK and ADME in Drug Discovery

A compound can work well against an isolated target yet fail to produce the expected effect in an animal study. It may not reach sufficient exposure, may be cleared rapidly or may distribute poorly to the tissue of interest. Understanding these possibilities is central to DMPK services.

DMPK stands for drug metabolism and pharmacokinetics. It examines how compounds are transformed and how their concentrations change over time in biological systems. ADME describes four related processes: absorption, distribution, metabolism and excretion.

Follow what happens after administration

Absorption concerns movement from the administration site into systemic circulation; intravenous administration bypasses this absorption step. Distribution concerns movement between blood and tissues.

Metabolism changes a compound through biochemical reactions, often involving enzymes. Excretion removes the parent compound or its metabolites from the body. These processes together influence exposure, but their importance varies by molecule and route of administration.

For an oral compound, poor exposure could reflect low solubility, limited absorption or extensive metabolism before reaching systemic circulation. The same observation can therefore have several explanations.

Use early experiments to narrow the possibilities

ADME services may include measurements of solubility, permeability, protein binding and metabolic stability. Different experiments answer different questions.

A metabolic stability assay can indicate how quickly a compound disappears in a specified test system. It does not directly establish the compound’s half-life in humans. Similarly, a permeability result offers information about movement across the model used, rather than a complete prediction of oral absorption.

The value comes from interpreting the studies together and recognising the limitations of each model.

Connect exposure with biological effects

Pharmacokinetic studies measure concentrations at selected times after dosing. Depending on the study, researchers can estimate exposure, clearance and other parameters.

Suppose two compounds show similar activity in a cellular assay, but one produces a stronger effect in vivo. Exposure data may help explain the difference. Conversely, an ineffective compound may have reached adequate concentrations, suggesting that the biological hypothesis or model needs closer examination.

Total blood or plasma concentration is not always the relevant measure. Unbound concentrations and exposure at the site of action can be important, depending on the target and programme.

Feed the findings back into chemistry

DMPK should inform compound design while chemical changes are still feasible. If a series is rapidly metabolised, metabolite identification may help identify vulnerable regions. Chemists can then explore modifications and test whether the changes improve the overall profile.

The team must watch for trade-offs. A modification that improves metabolic stability may reduce solubility or activity. No single assay can settle the choice.

Distinguish discovery support from a complete development package

Exploratory DMPK studies help rank compounds and explain behaviour. Preclinical development services may involve additional studies, documentation and quality requirements appropriate to the intended regulatory use. An early screening result does not replace that later work.

Jubilant Biosys offers in vitro and in vivo DMPK support. When planning DMPK services, sponsors should define the decision each study needs to support and how its findings will influence chemistry, biology or development planning.
